Effect of Storage Conditions on the Titer of Mycobacterial Phages


Abstract
Mycobacterial phages, suspended in saline and broth in the concentration of 1X109 per ml., were stored at various temperatures and for variable duration of time. The host cell was H37Rv and the media used for the culture were RVB and RVA. Comparison of the number of the phages, DS6A and AGI, survived the treatment was as follows.
1. The decrement of phage titer at lower than 4 C was less than 10-2 for 10 days and roughly 10-3 for three months, but when the storage temperature was higher than room temperature (15 C), the titer decrement was so sharp even at 10 days that the phage suspension was no longer useful in any phage typing work; needless mentioning about three month storage.
2. Broth seemed slightly better than physiological saline as the suspending medium for the mycobacterial phages.
3. DS6A phage seemed a little more sensitive to the storage conditions than the phage AG1.
